How to identify Shigella
1. Preparation of antigen
Shigella is not motivated and therefore does not have flagellar antigens. Shigella mainly contains bacterial (O) antigens. The bacterial O antigen can be divided into type and group specific antigens.
Agar cultures of 1.2% to 1.5% are generally used as antigens for slide agglutination tests.
Note 1: If some Shigella strains do not show agglutination due to the presence of the K antigen, they can be picked up in 1 mL of saline and made into concentrated broth. After boiling for 15 min to 60 min at 100°C to remove the K antigen, an examination.
Note 2: Shigella group D may be either a smooth strain or a rough strain, and there is no cross reaction with other Shigella antigens. Unlike the Enterobacteriaceae, the strains of Shigella sonnei rough may not self-condense. There is no K antigen in Shigella sonnei.
2 Agglutination reaction
Place two approximately 1cm x 2cm areas on the slide, pick a loop of the bacteria to be tested, place a 1/2 loop on the upper part of each area on the slide, and add 1 drop of antiserum to the lower part of the area. One drop of saline was added to the lower part of the area as a control. The bacterial colonies in the two areas were further ground into emulsions with sterile loops or needles. The slides were shaken and mixed for 1 min, and observed against a black background. If agglutinated particles appeared in the antiserum, and no coagulation occurred in the saline, the agglutination reaction was positive.
If agglutination occurs in saline, it is considered as self-condensation. At this time, other colonies on the same medium should be picked to continue the test.
If the biochemical characteristics of the test bacteria are in line with the biochemical characteristics of Shigella, and the serological test is negative, perform the test according to 5.5.1 Remark 1.
3 Serological typing
First, four types of Shigella polyvalent sera were used for examination. If agglutination was present, the respective groups of polyvalent sera were tested separately. The first experiment was carried out with group B multivalent serum of Shigella flexneri, and if it showed aggregation, it was checked separately with its group and type factor serum. If group B polyserum does not agglutinate, then test with group D Shigella sonnei sera, if present agglutination, then check with its phase I and II serum; if neither group B nor group D have agglutination , then use A group Shigella dysenteriae multivalent serum and 1 to 12 various types of factor serum examination, if the above three kinds of polyvalent serum does not agglutinate, can use C group Shigella spp. multivalent check, and further use 1 to 18 various types of serum tests.
